the genomic DNA was randomly fragmented using Megaruptor (Diagenode, USA). Subsequently, adapter preparation and ligation were performed using the Nanopore SQK-LSK 109 kit (Oxford Nanopore Technologies, USA). The ligated DNA library was then quantified using the Qubit 3.0 Fluorometer. Finally, the prepared sample was loaded onto Nanopore Flow cells R9.4 and sequenced on the PromethION platform. |
