Following the Iso-Seq protocol, cDNA synthesis and amplification were performed using the NEBNext Single Cell/Low Input cDNA Synthesis & Amplification Module. Subsequently, the PacBio SMRTbell Express Template Prep Kit 2.0 was utilized for sample processing, including adapter ligation and addition of SMRTbell sequences. Size-selective purification was then conducted using the ProNex® Size-Selective Purification System to remove low-quality and short fragment sequences, completing the preparation of the Iso-Seq library. |
