Tissues were collected for frozen sections. After quality control, tissue permeabilization, reverse transcription and two rounds of barcodes ligation were continuously conducted, then the reverse crosslinking were carried out. The lysate were purified and used for MyOne C1 beads binding to collecting cDNA and next template switch reaction. After amplification, the Novoprotein Tn5 enzyme were used to preduce random fragments. The 3 end products were amplified with illumina Nextera adapters and the libraries were sent for next generation sequencing. |
