DNA for each sample were extracted by Power Soil DNA Isolation Kit (Qiagen, Germany), then the V3-V4 region of the 16S rDNA was alpliedied by two-step PCR,Library was prepared following the protocol of the NEBNext® Ultra™ II DNA Library Prep Kit for Illumina® (New England Biolabs, USA). Finally, the amplicons were sequenced on Illumina Novaseq 6000 platform in pair-end 250 bp mode. |
