Briefly, dozens of oocytes and embryos had their zona pellucida removed and were subsequently washed with 0.1% BSA/PBS three times and lysed in TRIzol. Total RNA was extracted using Direct-zol RNA MicroPrep (Zymo Research) according to the manufacturer’s instructions and resolved in 12 μL nuclease-free water. The rRNA was removed using the Ribo-off rRNA Depletion Kit (Vazyme) and the libraries were constructed with the VAHTS Universal V8 RNA-seq Library Prep Kit for Illumina (Vazyme) following the manufacturer’s instructions. The prepared libraries were sequenced on the Illumina platforms. |
