2 g of young leaves of 2-month-old A. mongolicus was cross-linked with 40 mL of 2% formaldehyde at room temperature for 15 min, and then quenched by adding 4.324 mL of 2.5 M glycine. A Hi-C library was constructed according to the method described by Shi et al. (2019). DNA was digested overnight with a 4-bp cutter restriction enzyme (400 units of MboI) at 37C on a rocking platform. Hi-C libraries were amplified by 1012 cycles of PCR. |
