Ten million primary macaque neuronal cells cultured at day 15 were crosslined, lysed to permeabilize cell membrane, and treated with AluI to digest DNA. The DNA ends were A-tailed and DNA ends in the proximity were then ligated to the same biotinylated linkers. After sonication, 20 microgram of CTCF antibody were applied to capture CTCF binding chromatin. The ChIP DNAs were decrosslinked, purified, and Tn5 tagmented, and then the DNAs with biotin were captured on streptatvidin beads. The DNAs on beads were then subjected to index PCR for libarary construction. |
