EBJC cells were crosslinked and sonicated using the Covaris truChIP-seq chromatin shearing kit (Covaris, Cat. No. 520154). Sixty µg sheared chromatin (corresponding to ten million cells) was incubated overnight with five µg antibodies against H3K27ac coated on protein-G beads. The enriched DNA was eluted from the protein-G beads, column purified, and then end-repaired, ligated to index adapters, and PCR-amplified with ultralow V2 DNA-seq library preparation kit (Nugen, Cat. No. 0344NB) to construct libraries for sequencing. |
