The purified cells were next resuspended in PBS, loaded onto a 10x Chromium microfluidics system with Single Cell 5’ Library Gel Bead Kit according to the manufacturer’s guidelines. Completed libraries were sequenced on Illumina Novaseq 6000 platforms at a targeted mean read depth of 50,000 reads per cell from total gene expression libraries.
