Young and fresh leaves were fixed in a nuclei isolation buffer with 2% formaldehyde, then cross-linked DNA was digested with 100 units of DpnII (New England Biolabs, USA). The digested fragments were biotinylated with biotin-14-dCTP and ligated using T4 DNA polymerase (New England Biolabs). The ligated DNA was enriched, sheared into 300- to 600-bp fragments, blunt-end repaired, and further processed. |
