For quality-assured RNA (or polyA+ RNA), the first cDNA strand is synthesized using the SMARTer PCR cDNA Synthesis Kit. When SMARTScribe Reverse Transcriptase reaches the 5’ end of the mRNA, terminal transferase activity adds several extra bases to the 3’ end of the cDNA. These bases serve as binding sites for oligonucleotides, extending the first cDNA strand to the oligonucleotide’s end and generating a universal primer sequence to facilitate second-strand synthesis and PCR amplification. |
