Genomic DNAs from fruitfly S2 cell line were digested with HgaI (ER1901, Thermo Fisher Scientific, Waltham, US), and the resulting 5-bp nicks were filled using a polymerase reaction with dMTPs (M representing Z/T/C/G). The repaired DNA fragments contain mostly As and a limited number of Zs at their terminal(s) and were sequenced using PacBio CCS on Revio platform. |
