Biological specimens including bronchoalveolar lavage fluid (BALF) or sputum, were collected from enrolled patients suspected of having tuberculosis. A minimum of 800 μL of each sample was obtained. Microbial DNA was extracted using nucleic acid extraction and purification reagents from GenK magnetic nead-based DNA/RNA extraction kit. Extracted DNA was subjected to targeted multiplex-PCR using a pre-designed primer panel (covering targeted regions of 64 microbial species and 158 drug resistant sites of MTB) to enrich pathogen-specific genomic fragments. This step utilized the GenK Tuberculosis Infection Pathogen Nucleic Acid Detection Kit (Multiple amplification method). After the targeted enrichment, adapters were added to the DNA to complete the library construction. |
