The RNA was extracted by RNA extraction kit. Processing total RNA using either mRNA enrichment (capturing polyA-tailed mRNA with Oligo dT beads) or rRNA removal (digesting rRNA-DNA probe hybrids with RNaseH/DNaseI). Fragmenting RNA and synthesizing double-stranded cDNA using random N6 primers. End-repairing DNA with phosphorylation, adding 'A' overhangs, and ligating bubble-shaped adapters with 'T' overhangs. Amplifying ligated DNA through PCR and circularizing single-stranded products using bridge primers. Performing sequencing after obtaining single-stranded circular DNA libraries. |
