The Hi-C library preparation involved fixing fresh O. corymbosa young leaves in 2% formaldehyde. The fixed tissues were homogenized and centrifuged to isolate nuclei. The cross-linked chromatin was digested with DpnII, labeled with biotin, and ligated with T4 DNA ligase. After de-crosslinking, the DNA was purified and fragmented into 300 to 500 bp segments. Fragments containing interaction sites were enriched using streptavidin magnetic beads to construct the Hi-C library. |
