Nucleic acid extraction was performed using a column-based method in which functional groups with an affinity for nucleic acids were fixed onto a centrifugal column. By adding different lysis and washing reagents followed by repeated centrifugation, nucleic acids were separated from impurities, resulting in purified nucleic acids. The library was then constructed using a specialized pathogen DNA library preparation kit. In this process, transposase was used to fragment the genome into appropriately sized segments. Sequencing adapters compatible with the Illumina™ sequencing platform were attached to both ends of the fragments, and specific regions were captured using custom or specific primers. Subsequently, PCR amplification was performed to increase the library quantity to meet sequencing requirements. |
