Total RNA was isolated and purified from root tissue samples using TRIzol reagent, with >1 μg of total RNA meeting the requirements for downstream experiments. PolyA-tailed mRNA was specifically captured using oligo(dT) magnetic beads. The captured mRNA was fragmented at elevated temperature and subjected to paired-end sequencing (PE150) on an Illumina NovaSeq™ 6000 platform (LC Bio Technology Co., Ltd., Hangzhou, China) following standard operating procedures. |
