The samples were dissociated and nucleated. The Gel Beads containing barcode information were combined with the mixture of cells and enzymes to form GEMs (Gel beagle-in-emulsions). The gel beads dissolved to release the capture sequences containing Barcode sequences, reverse transcribed cDNA fragments, and labeled the samples. The gel beads were crushed and the oil droplets were broken up, and PCR amplification was performed using cDNA as a template. Mix all the products of GEMs to construct a sequencing library |
