Prior to conducting Single Molecule Real Time (SMRT) sequencing, high-quality genomic DNA underwent stringent quality control. The genomic DNA was fragmented into suitably sized large fragments, and then damage repair, adapter ligation, and fragment selection were performed to construct a PCR-free SMRT bell library.
