Total RNA was isolated from the peri-injury regions at 6 hours post the final treatment, following three consecutive days of treatment, using NcmSpin Cell/Tissue Total RNA Kit (NCM Biotech, Suzhou, China) in strict accordance with the manufacturer’s recommended protocol. RNA quality was subsequently quantified using a NanoDrop ND-2000 spectrophotometer (NanoDrop Technologies, Wilmington, DE, USA) and further characterized for integrity using an Agilent 5300 Bioanalyzer (Agilent Technologies, Santa Clara, CA, USA). Only high-quality RNA sample (OD260/280 ≥ 1.95, RQN ≥ 6.5, 28S:18S ≥ 1.0, > 1μg) was used to construct sequencing library. |
