To delineate the dynamic regulatory network of the viral infection-host response, we infected Huh7 (immune-competent) and Vero-E6 cells (immune-deficient30) with the SARS-CoV-2 strain. Both cell lines are highly susceptible to SARS-CoV-2 infection due to their expression of the viral entry receptor ACE2. Huh7 cells were cultured under two conditions, with and without SARS-CoV-2 infection. Vero-E6 cells were cultured under four conditions: mock control (untreated), infected by SARS-CoV-2, treated with remdesivir (RDV), and treated with both RDV and SARS-Cov-2. The total RNAs were individually extracted from each sample at 3 hours post infection (h.p.i.), 18 h.p.i. and 24 h.p.i. to simulate the early, middle and late stages of viral infection, and subjected to Poly(A) RNA enriched library constructions before deeply sequenced on Illumina Novaseq system. |
