Fecal samples were processed for viral metagenomic sequencing as follows: samples were suspended in 10 mL of sterile PBS, vortexed, and centrifuged at 7000 g for 10 min. Supernatants were filtered through a 0.22-um sterile membrane and treated with 6% (w/v) polyethylene glycol 6000 and 0.2 M NaCl to precipitate particles for 2 h. After centrifugation, the pellet was resuspended in 200 uL of nuclease-free water and treated with DNase I for 10 min to remove free DNA. Viral nucleic acids were extracted using the Tiangen Viral DNA/RNA Extraction Kit according to the manufacturers protocol. Sequencing libraries were prepared with the Illumina TruSeq DNA Sample Preparation Kit, following the manufacturers protocol, and sequenced on an Illumina NovaSeq 6000 platform, generating 150-bp paired-end reads. |
