The cell suspension is injected into the SCOPE-chip™ microfluidic chip, where single cells are separated based on the principle of the "Poisson distribution." Cells fall into specially designed chip micro-wells under the influence of gravity, ensuring that only one cell falls into each micro-well. Subsequently, millions of magnetic beads carrying unique cell tags (Cell Barcodes) are added to the chip micro-wells, ensuring that only one magnetic bead falls into each micro-well. After cell lysis, the magnetic beads with unique cell tags (Barcodes) and molecular tags (UMI) capture mRNA by binding to the poly(A) tail of mRNA, labeling both the cells and the mRNA |
