The pituitary tissue was dissected from E18.5 embryos and washed with ice-cold PBS, then incubated in 10 U/ml papain for 20 min at 37 C, 5 percent CO2 in air and pipetted gently every 10 min. The digestion was halted by adding DMEM containing 10 percent FBS and 100 U/ml DNase I to reduce viscosity. Single-cell suspensions were loaded onto a Chromium chip to generate single-cell gel bead-in-emulsions (GEMs) for capturing 500 to 10000 single cells per reaction. The single-cell 3 prime library was constructed using Chromium Single Cell Reagent Kits v3 (10x Genomics) following the manufacturer's instructions. The DNA libraries were sequenced on an Illumina NovaSeq 6000. |
