Sequencing libraries are prepared with 500 ng input DNA using the KAPA Hyper Library Preparation Kit (PCR-free). Covaris shearing targets 350 bp fragments. Libraries are multiplexed in pools of 20 and each pool is sequenced on 2 1/2 flow cells of HiSeq X.
