Fluidigm 2 Step Access Array Amplification and High Throughput Sequencing. Library construction and sequencing were performed at the Roy J. Carver Biotechnology Center, University of Illinois at Urbana-Champaign. Approximately 2 ng of DNA was used for amplification of the targeted regions in the Fluidigm Access Array (Fluidigm, CA) as previously described (Muturi, et al., 2016). The pooled amplicons were sequenced on one lane with 2 x 250nt paired-reads on a HiSeq 2500 with a Rapid sequencing kit version 2 (Illumina, CA). Fastq files were generated and demultiplexed with the bcl2fastq v2.17.1.14 Conversion Software (Illumina). |
