High molecular weight (HMW) DNA has been extracted from young leaves tissue using the protocol of Doyle and Doyle (1987) with minor modifications; sheared to achieve an appropriate size range for constructing a Pacbio Hi-Fi sequencing library, constructed following manufacturer protocols using SMRTbell Express Template Prep kit 2.0. The final library has been size selected, then quantified, size checked, and sequenced on PacBio Sequel II using standard protocols from the manufacturer. |
