Library prep uses 500 ng input DNA sheared by Covaris followed by a double size selection step using different ratios of AMPure XP beads. It uses KAPA Hyper PCR-free library reagents (KK8505, KAPA Biosystems, Inc.) and barcoded adapters to create PCR-free libraries. One sample is loaded per HiSeq X lane.
