PTENP1, phosphatase and tensin homolog pseudogene 1 is a tumor suppressor gene and undergoes genomic deletion in human melanoma.

Characteristics

PTENP1 is 3932 bp long non-coding RNA, located on human chromosome 9p13.3 (HGNC). PTENP1 is highly homologous to PTEN, with only 18 mismatches throughout the coding sequence [1].

Function

Functions of long non-coding RNA PTENP1[2]

PTENP1 regulate the expresssion of tumor supressor gene PTEN [1][3]. PTENP1 3'UTR can increase PTEN expression by binding to microRNAs that downregulate PTEN expression. Thus, PTENP1 plays an important role in cancer biology, restricting cell proliferation by acting as a microRNA decoy for the tumor suppressor PTEN [1].

The overexpressed PTENP1 repressed the oncogenic PI3K/AKT pathway and elicited pro-death autophagy via sequestering miR-17, miR19b and miR-20a in vitro, and inhibited the hepatocellular carcinoma (HCC) tumor growth in vivo, which was accompanied by enhanced apoptosis, autophagy and dampened angiogenesis/neovasculature maturation [3].

PTENP1 along with two other lncRNA CUDR, and LSINCT-5 was identified as a potential diagnostic marker for gastric cancer (GC) that distinguished between early-stage GC patients and healthy subjects [4].

Histone acetylation is a key factor in controlling lncRNA BANCR expression [1].
