GSC-DT, is a conserved long noncoding RNA and a key regulator of human and mouse definitive endoderm (DE) differentiation.

Annotated Information

Name

GSC-DT: GSC divergent transcript (HGNC nomenclature), DIGIT [1].

Characteristics

GSC-DT is two exon transcript of 2226 bp long non-coding RNA (GenBank), located on chromosome chr14q32.13 (HGNC).

Function

Loss of GSC-DT expression inhibits definitive endoderm differentiation. [1]

GSC-DT regulates Goosecoid (GSC) expression to control definitive endoderm differentiation of both human and mouse embryonic stem cells [1].

Goosecoid (GSC) expression is regulated by GSC-DT in trans, and the defect in definitive endoderm (DE) differentiation present in GSC-DT-deficient hESCs can be rescued by induction of GSC. [1].

Expression

GSC-DT is expressed during embryonic development [1].

Regulation

GSC-DT regulation through SMAD3. [1]

GSC-DT regulated by an enhancer bound by SMAD3 following activin signaling during endoderm differentiation [1].
