Bidirectional promoter with HOXA11. Multiple splice isoforms are present which share a common 3' exon ([http://www.ncbi.nlm.nih.gov/pubmed/7789268 Hsieh-Li (1995)], [http://www.ncbi.nlm.nih.gov/pubmed/9745033 Potter (1998)]), only some overlap HOXA11 [http://www.ncbi.nlm.nih.gov/pubmed/9745033 (Potter (1998)]). Annotated human isoform is ~1.6kb. A number of isoforms including ~1.4, ~1.9, ~2.9 and ~3.2kb transcripts have been identified in mouse [http://www.ncbi.nlm.nih.gov/pubmed/7789268 (Hsieh-Li (1995))].

===Function===
 +
Due to the inverse or complementary expression pattern between Hoxa11 and HOXA11as mRNA expression levels, in the developing limbs [http://www.ncbi.nlm.nih.gov/pubmed/7789268 (Hsieh-Li (1995))] and adult endometrium [http://www.ncbi.nlm.nih.gov/pubmed/12050232 (Chau (2002))], HOXA11as has been proposed to negatively regulate Hoxa11 mRNA levels [http://www.ncbi.nlm.nih.gov/pubmed/12050232 (Chau (2002))].

===Expression===
 +
More highly expressed than HOXA11 in mouse development [http://www.ncbi.nlm.nih.gov/pubmed/7789268 (Hsieh-Li (1995))]. Expressed in developing limbs where it showed a complementary expression pattern to HOXA11 [http://www.ncbi.nlm.nih.gov/pubmed/7789268 (Hsieh-Li (1995))]. Also expressed in fetal kidney [http://www.ncbi.nlm.nih.gov/pubmed/9745033 (Potter (1998))],  placenta, adult colon, muscle and prostate ([http://www.ncbi.nlm.nih.gov/pubmed/17185360 Sessa (2007)], [http://www.ncbi.nlm.nih.gov/pubmed/17445766 Sasaki (2007)]). Shows collinear regulation of expression like HOX PTC genes [http://www.ncbi.nlm.nih.gov/pubmed/17445766 (Sasaki (2007))]. Expressed in same cell types as HOXA11 in the uterus. HOXA11as was dynamically expressed in the uterus during the menstrual cycle, with a peak in the endometrial late-proliferative phase, showing a pattern inverse to HOXA11 [http://www.ncbi.nlm.nih.gov/pubmed/12050232 (Chau (2002))]. HOXA11as levels were down-regulated by progesterone in primary uterine stromal cells, which correlated with HOXA11 up-regulation in the same cells [http://www.ncbi.nlm.nih.gov/pubmed/12050232 (Chau (2002))].

Some HOXA11as isoforms have long ORFs, however these ORFs are not conserved between human and mouse (despite high nucleotide sequence conservation) ([http://www.ncbi.nlm.nih.gov/pubmed/7789268 Hsieh-Li (1995)], [http://www.ncbi.nlm.nih.gov/pubmed/9745033 Potter (1998)]). So while HOXA11as is thought to be noncoding, a possible role for HOXA11as encoding a protein cannot be completely discounted.

Annotated Information

Name

Hoxa11as

Characteristics

Bidirectional promoter with HOXA11. Multiple splice isoforms are present which share a common 3' exon (Hsieh-Li (1995), Potter (1998)), only some overlap HOXA11 (Potter (1998)). Annotated human isoform is ~1.6kb. A number of isoforms including ~1.4, ~1.9, ~2.9 and ~3.2kb transcripts have been identified in mouse (Hsieh-Li (1995)).

Function

Due to the inverse or complementary expression pattern between Hoxa11 and HOXA11as mRNA expression levels, in the developing limbs (Hsieh-Li (1995)) and adult endometrium (Chau (2002)), HOXA11as has been proposed to negatively regulate Hoxa11 mRNA levels (Chau (2002)).

Expression

More highly expressed than HOXA11 in mouse development (Hsieh-Li (1995)). Expressed in developing limbs where it showed a complementary expression pattern to HOXA11 (Hsieh-Li (1995)). Also expressed in fetal kidney (Potter (1998)), placenta, adult colon, muscle and prostate (Sessa (2007), Sasaki (2007)). Shows collinear regulation of expression like HOX PTC genes (Sasaki (2007)). Expressed in same cell types as HOXA11 in the uterus. HOXA11as was dynamically expressed in the uterus during the menstrual cycle, with a peak in the endometrial late-proliferative phase, showing a pattern inverse to HOXA11 (Chau (2002)). HOXA11as levels were down-regulated by progesterone in primary uterine stromal cells, which correlated with HOXA11 up-regulation in the same cells (Chau (2002)).

Conservation

Sequence highly conserved (Potter (1998)), found throughout mammals.

Misc

Some HOXA11as isoforms have long ORFs, however these ORFs are not conserved between human and mouse (despite high nucleotide sequence conservation) (Hsieh-Li (1995), Potter (1998)). So while HOXA11as is thought to be noncoding, a possible role for HOXA11as encoding a protein cannot be completely discounted.
