Annotated Information

Name

WT1-AS: WT1 (Wilms' tumor 1) antisense transcript.

Characteristics

0.4 - 3kb, spliced and present in multiple splicing isoforms (Dallosso (2007)), as well as a protein coding isoform (WIT1) transcribed from a downstream promoter (Eccles (1994),Campbell (1994)). Transcribed antisense to WT1 from intron 1 or from the same promoter region (bidirectional) of WT1, corresponding to a differentially methylated regulatory region (Eccles (1994),Malik (1995),Malik (2000)).

Function

WT1-AS transcripts are exported to the cytoplasm and form heteroduplexes with WT1 mRNA, with a proposed regulatory role in WT1 expression via RNA:RNA interactions (Dallosso (2007)).

Expression

Imprinted in humans (but not in mouse), with monoallelic (paternal) expression in normal kidney (Malik (2000),Dallosso (2004),Dallosso (2007)). Co-regulation with WT1 during mouse kidney development, and co-expression in mouse and human tissues, including fetal spleen, fetal kidney, and normal kidney (Dallosso (2007)). Abnormal splicing of WT1-AS has been reported in acute myeloid leukaemia (AML) samples (Dallosso (2007)).

Conservation

WT1 has a positionally conserved antisense/bidirectional transcripts in mouse and humans, although WT1-AS RNAs do not show extensive sequence conservation between these species, except at their 5' ends, which may extend into WT1 exon 1 (Dallosso (2007)).
