Characteristics
HNF1A-AS1 is a 2455-nucleotide single-exon gene located at chromosomal band 12q24.31 and it is also a bidirectional IncRNA corresponding to the hepatic nuclear factor 1 alpha gene (HNF1A, also known as transcription factor 1 or TCF1), whose transcriptional start site occurs approximately 5 kb 5′ to that of HNF1A-AS1 [1].
Function
HNF1A-AS1 is involved in the promotion of EAC cell proliferation and anchorage-independent growth, two cancerous processes in epithelial cells[1]. HNF1A-AS1 involves mechanisms relevant to the promotion of EAC cell migration and/or invasion [1]. At least one HNF1A-AS1-responsive pathway likely involves chromatin/nucleosome assembly during the G1/S transition [1]. HNF1A-AS1-mediated promotion of EAC cell proliferation and survival appears to be mediated by modulation of the G1-S checkpoint, rather than by apoptosis [1].
Regulation
H19 may represent an important downstream effector of HNF1A-AS1, potentially mediating this IncRNA’s effects on tumour growth [1].
Evolution
HNF1A-AS1 is evolutionally poorly conserved , yet widely expressed in the aerodigestive tract [1].
Expression
There is a significant positive correlation between HNF1A-AS1 and H19 expression in primary EACs [1].
HNF1A-AS1 is significantly up-regulated in lung adenocarcinoma tissues compared with corresponding non-tumor tissues, and its expression level is significantly correlated with TNM stage, tumor size, and lymph node metastasis [2].
