Tmevpg1: Theiler's murine encephalomyelitis virus persistence candidate gene 1
Characteristics
Found within the Tmevpg3 genetic locus which controls Theiler's virus persistance/ load (Vigneau (2001)). Promoter contains binding sites for E2A and the Ets family of transcription factors. Suggesting regulation by transcription factors involved in the immune system (Vigneau (2001)). Spliced with 5 or 6 exons depending on isoform/ species (Vigneau (2001)).
Function
It has been speculated there may be a regulatory regulationship between Tmevpg1 and Ifng to explain their reciprocal expression (Vigneau (2003)).
Expression
In resistant mice Tmevpg1 is constitutively expressed in CD4+ and CD8+immune cells in the spleen and thymus and upon infection, by immune cells infiltrating into the CNS, however no Tmevpg1 expression was seen in susceptible mice (Vigneau (2001), Vigneau (2003)). Found in peripheral blood lymphocytes in humans with expression in NK+ cells, CD4+ and CD8+ T-lymphocytes (Vigneau (2001), Vigneau (2003)). In both mouse and humans Tmevpg1 was down-regulated upon immune cell activation with a reciprocal up-regulation in interferon gamma (Ifng) expression (Vigneau (2003)).
Conservation
Present in mouse and human. Sequence conservation is limited to the promoter, first exons and its splice site, otherwise the transcripts are poorly conserved and can contain repeat elements (Vigneau (2001)).
Misc
The Tmevpg3 genetic locus was found to control persistence of Theiler's virus in the central nervous system, along with the H-2D gene, this QTL locus was found to be near but exclude the interferon gamma (Ifng) gene (Bihl (1999)).
