Characteristics
Multiple Tug1 isoforms, with a main 6.7kb transcript in mouse. Spliced [1].
Function
Required for the proper formation of photoreceptors in the developing rodent retina [1]. Downregulation in developing retina leads to decreased retinal transcription factor expression and increased apoptosis [1]. TUG1 expression appears to be activated by p53 upon DNA damage, with many p53 binding sites in the Tug1 promoter [2][3]. Tug1 associates with PRC2 (polycomb repressive complex 2) and represses a number of cell cycle genes [3].
Expression
Expressed in the developing retina and brain [1]. Expressed in adult tissues with highest expression in cortex [1]. Expression levels are elevated in the caudate nucleus of patients suffering from the trinucleotide repeat neurodegenerative condition, Huntington's disease (Johnson (2012)). Localises to both the nucleus and cytoplasm [3]. Transcript was classified as unstable with a half-life >4 hr in human Hela cells (Tani (2012)). Similar results were found in human B cells and mouse 3T3 cells (Friedel (2009), Clark (2012)).
Conservation
Mammals - identified in mouse, rat, cow, dog and human [1]. Present in two locations in rat, with one an apparent processed pseudogene.
Misc
Nb: Mouse NRED accession is specific to one of two Tug1 isoforms.
