Characteristics

LINC00853 is a long intergenic noncoding RNA also known as ncRNA-a4 which does not overlap annotated protein-coding regions. It has chromosomal location at 1p33. In vitro translation assay confirmed that this ncRNA does not yield any protein product [1].

Function

LINC00853 plays a major role in transcriptional activation of the gene CMPK1 which is over 150 kb downstream of it [1].

Regulation

siRNAs specific to ncRNA-a4 reproducibly reduced its levels by about 50 percent [1].

Expression

Expression of LINC00853 was reliably detected in Jurkat cells, however, it was not expressed at a detectable level in MCF7 cells in a qPCR study. Importantly, reduced levels of LINC00853 resulted in a consistent and significant decrease in the level of the gene CMPK1 but no change was detected in the other protein-coding genes surrounding LINC00853. Depletion of LINC00853 resulted in a partial decrease in transcriptional enhancement suggesting that other DNA elements may also contribute to increased transcription [1].
