In univariate analysis, we observed that patients who had a history of primary refractory disease showed a worse response to 7 x19 CAR T cell therapy, suggesting that certain patient-related factors may also affect the clinical efficacy of 7 x 19 CAR T cells. Considering that metabolic reprogramming is imperative for CAR T cell function and high metabolic demands of cancer cells in patients may compromise the anti-tumor activity of CAR T cells30,31, we investigated the possibility that metabolic status of R/R LBCL patients may affect the efficacy of 7 x 19 CAR T cell therapy. Metabolome profiles in 20 responders and 6 nonresponders before or at 1, 2, and 4 weeks post-infusion were analyzed using liquid chromatography-tandem mass spectrometry (LC-MS/MS). |
