Description | AT2 cells were stimulated in the culture medium with different anti-tuberculosis drugs (10 μg/mL rifampicin, 10 μg/mL ethambutol, 10 μg/mL levofloxacin, 10 μg/mL moxifloxacin) for 24h and prepared for metabolomics analysis. As we described before [PMID: 32059792], AT2 cells were added 1 mL of methanol and 20 µL of 4 µg/mL L-2- chloro-phenylalanine as internal standard. Chloroform was added and the cells were extracted by ultrasonication for 20 minutes. The supernatant was transferred to glass vials to evaporate the components. GC/MS data was processed by MS-DIAL software. In each sample, all peak signal intensities were segmented and normalized according to the internal standards with RSD greater than 0.1 after screening. The matrix was imported to carry out Principle Component Analysis (PCA) to observe metabolic differences between groups and the stability of the whole analysis process. |
