| Description | Peptides were separated by a 60 min gradient elution at a flow rate 0.3 μL/min either with Thermo easy nLC 1000 system coupled with a Thermo LTQ Orbitrap Elite mass spectrometer. The analytical column was a homemade fused silica capillary column packed with C18 resin (5 μm for perfusate or 3 μm for human heart tissue samples). Mobile phase A consisted of 0.1% formic acid, and mobile phase B consisted of 0.1% formic acid in acetonitrile. The mass spectrometer was operated in data-dependent mode with a full MS scan (375-1600 M/z) in FT mode at a resolution of 120000 followed by CID MS/MS scans on the 15 most abundant ions in the initial MS scan. |
