Description | The process includes sample pretreatment, metabolite extraction, metabolite derivatization, LC-MS detection, data pretreatment, and statistical analysis. Muscle tissue metabolomics was analyzed using the ACQUITY UPLC I-Class system coupled with the VION IMS Q-Tof Mass Spectrometer System (Waters, Milford, MA), covering both ESI positive and negative ion modes. The capillary voltage, electrospray ionization (EP), and collision energy (CE) were set to 1.0 kV, 40 V, and 6 eV, respectively. The source temperature and desolvation temperature were maintained at 120°C and 500°C, respectively, with a desolvation gas flow rate of 900 L/h. Data were collected over a 13-minute analysis period, with a scan range of 50 to 1000 m/z, using a scan time of 0.1 seconds and an interscan delay of 0.02 seconds. |
