Description | For proteomic analysis of IP samples, protein digestion was carried out using a modified FASP method. In brief, 100 µg of protein underwent reduction, alkylation , and washing to eliminate stains and SDS. The samples were then digested overnight with trypsin at 37°C, and the resulting peptides were collected and diluted with 0.1% formic acid for nanoLC-MS analysis. NanoLC separation was performed using a Waters (Milford, MA, USA) nanoAcquity nanoHPLC with a gradient elution method. Nanospray ESI-MS was conducted on a Thermo Q-Exactive high-resolution mass spectrometer (Thermo Scientific, Waltham, MA, USA). |
