| Description | The protease inhibitor PMSF and the phosphatase inhibitor were added to 100mg of gastric tissue sample, so that the final concentration was 1mM. Add 1.5 times acetone per milliliter of TRIzol system, mix well, and precipitate overnight at -40. Centrifuge at 4, 12000 rpm for 10min, discard the supernatant; Add 500uL 90% acetone at -40 for 30min-1h. Centrifuge at 4, 12000 rpm for 10min, discard the supernatant. Add 100 μL SDS lysate, add phosphatase inhibitor and PMSF to make the final concentration of 1mM, dissolve at 50°C for 1-2h.The solution was centrifuged at 4,12000rpm for 10min, and the supernatant was obtained. The supernatant is the total protein solution of the sample, and the protein concentration is determined and stored at -80°C. |
