Transcription of GUS from a GA-regulated α-amylase promoter was measured relative to expression of LUX (GUS:LUX ratio) driven by a constitutive ubiquitin promoter (B). Half-grains were incubated for 24 h without hormone (−GA) or with GA and the ratio of GUS-to-LUX expression determined in the presence and absence of the antisense construct.
