Annotated Information

Function

This gene encodes a novel CCHC-type zinc finger pronein called OsZFP6. OsZFP6 is structurally and functionally conserved, mainly localised in nucleus, and can play an important role in stress tolerance responses to salt (NaCl), alkali (NaHCO3), and H2O2 treatment in rice. Functional analysis also showed overexpression of this gene in transgenic Arabidopsis and yeast to NaHCO3 and H2O2.

Construction

The OsZFP6 gene is a single-copy gene in the rice genome. It is 1559 bp in length with a 458 bp 5' UTR and a 186 bp 3' UTR. The ORF encodes a 304 amino acid with a molecular weight of 33.6 kDa and an isoelectric point of 7.35. Based on structural properties indicated by the SMART programs, the predicted protein contains two conserved domains: a Pfam retrotrans-gag domain from amino acid 48 to 146 and a ZnF C2HC domain from amino acid 216 to 232.

Expression

Using the online analysis tool Psort to predict the subcellular localisation of rice OsZFP6, OsZFP6 was localised to the nucleus with a 73.9% probability. To validate the predicted result, a PBI121-OsZFP6:GFP plasmid was transformed into onion epidermal cells. After culture for 18-24 h in the dark, green fluorescence was observed in the nucleus (as shown in figure), suggesting the OsZFP6 protein, driven by the 35S promoter, was localized in the nucleus. Tissue distribution of OsZFP6 gene expression is still unknown yet.

Evolution

The CysCysHisCys (CCHC)-type zinc finger domains contain the sequence C-X2-C-X4-H-X4-C, where X can be any amino acid, and the numbers indicate the number of residues. This domain is conserved in Arabidopsis, yeast and rice, as shown in table.
