Function

OsAMT1;1(Os04g0509600)is one of the OsAMT1 family members which contain three genes(OsAMT1;1,OsAMT1;2 and OsAMT1;3).OsAMT1;1 has ammonium transporters activity.The amino acid sequences of the OsAMT1;1, OsAMT1;2 and OsAMT1;3 share high sequence similarity to each other and to other AMT1-type ammonium transporters previously identified from other plant species[1].All three isolated OsAMT1 genes encode functional ammonium transporters.OsAMT1;1 responds to ammonium in a comparable time and concentration-dependent manner,although absolute changes in transcript accumulation were less pronounced.The first step in nitrogen assimilation by plants is the uptake of the nitrate and ammonium into root cells from the soil solution. Since all three isolated transporter genes werefunctional in yeast, their translation products are thought to primarily contribute to the uptake of ammonium into the root.Because exclusive supply of ammonium is harmful, the ammonium taken up by the OsAMT1s is probably assimilated into glutamine through the reaction catalyzed by glutamine synthetase (GS) via glutamate through the reaction by glutamate synthase (GOGAT), designated as the concerted GS/GOGAT cycle[2].

Expression

OsAMT1;1 expression was constitutive in shoots and promoted by ammonium in roots;OsAMT1;2 expression was root specific and ammonium inducible,while OsAMT1;3 was also expressed specifically in roots but was repressed by nitrogen. These results implied feedback regulation, i.e. positive and negative control mechanisms for AMT1 genes in rice.It has been suggested that the cellular nitrogen status controls nitrogen and carbon metabolism as well as nitrogen absorption in roots[1][3].
