OsRDCP1 was induced by drought stress. Overexpression of OsRDCP1 increased tolerance to drought stress in rice.
Annotated Information
Function
Figure 1. Protein In vitro self-ubiquitination analysis of OsRDCP1.
OsRDCP1 exhinit self-ubiquitination activity in vivo. MBP-OsRDCP1 produced high-molecular-mass smear ladders in a time-dependent manner ( Fig. 1A). In contrast, no ubiquitination was detected when the E1, E2, ATP, or Ub was excluded from the reaction mixture(Fig. 1).
Figure 6. Overexpression of OsRDCP1 increased tolerance to drought stress in transgenic rice plants.
35S:OsRDCP1 transgenic lines exhibited considerably
less visual symptoms of drought stress. After 15 days
of drought stress, these transgenic lines appeared
healthy and showed reduced desiccation damage
relative to the wild-type and osrtbp1 plants. while
no significant phenotypic differences were detected
between the wild-type and osrtbp1 plants.
Homologues
Figure 2. Protein Identification of five homologous rice RING E3 OsRDCP family members.
These five rice OsRDCPs (O. sativa RING domain-containing proteins) paralogs
possess a single RING motif in their N-terminal
regions and a putative membrane-anchoring domain in
their C-terminal regions.The predicted OsRDCP
paralogs were 25–79% identical to each other at the
amino acid level .
among the five paralogs, OsRDCP1 was significantly induced by drought stress in both
shoot and root tissues, while the other OsRDCP
members were constitutively expressed ( Fig2). The
level of the OsRDCP4 transcript was the highest in
rice seedlings.
Expression
Figure 3. RNA gel blot analyses of OsRDCP1 mRNA. Light-grown 2- or 4-week- old rice plants .
OsRDCP1 transcript (approximately
1.2 kb) levels were significantly augmented in
response to 5–30% water loss ( Fig. 3C). This
increase was seen in both shoots and roots with
similar degrees of induction. Significant induction
of OsRDCP1 transcripts was also detected following
cold stress (2, 6, and 24 h at 4 °C) ( Fig. 3D). In
contrast, expression of OsRDCP1 was unaffected by
high salinity (150 mM NaCl) for at least 24 h.
Evolution
Figure 4. Phylogenetic relationship of RING Ub-ligase homologs from rice, Arabidopsis , hot pepper, and humans .
The coding region of OsRDCP1
is 762 bp and encodes 253 amino acids with a
molecular mass of 28.1 kDa and a calculated pI of
7.2. Full-length OsRDCP1 is 37%, 29%, and 31%
identical to hot pepper CaRma1H1 [12], Arabidopsis
AtRma1, and human RING protein HsRma1, respectively (
Fig. 4). In addition, the RING domain of OsRDCP1 is
48–78% identical to those of hot pepper,
Arabidopsis, and human RING proteins.
You can also add sub-section(s) at will.
