Sequencing results confirmed that the <i>OsZFP6</i> sequence was identical to Os02g0552700. The <i>OsZFP6</i> gene is a single-copy gene in the rice genome. It is 1559 bp in length with a 458 bp 5' UTR and a 186 bp 3' UTR. The ORF encodes a 304 amino acid with a molecular weight of 33.6 kDa and an isoelectric point of 7.35. Based on structural properties indicated by the SMART programs, the predicted protein contains two conserved domains: a Pfam retrotrans-gag domain from amino acid 48 to 146 and a ZnF C2HC domain from amino acid 216 to 232.

Using the online analysis tool Psort to predict the subcellular localisation of rice OsZFP6, OsZFP6 was localised to the nucleus with a 73.9% probability. To validate the predicted result, a PBI121-<i>OsZFP6</i>:<i>GFP</i> plasmid was transformed into onion epidermal cells. After culture for 18-24 h in the dark, green fluorescence was observed in the nucleus (as shown in figure), suggesting the OsZFP6 protein, driven by the 35S promoter, was localized in the nucleus.
