OsWRKY30 encodes a protein named WRKY30 transcription factor, which is a member of Group Ia of rice WRKY family with two DNA-binding domains, designated the WRKY domain, containing a highly conserved WRKYGQK residue as well as a novel zinc finger motif. Both the conserved elements of the domain are necessary for high binding affinity of WRKY30 to a canonical W-box containing a sequence of TTGACC/T [1][2].

WRKY30 is rapidly induced by pathogen M. grisea infection, bacterial pathogen Xanthomonas oryzae pv. oryzae (Xoo) and related phytohormones SA and JA [2]. OsWRKY30 functions as a positive regulator of rice disease resistance via an SA signaling pathway. It can up-regulate the SA-responsive gene OsWRKY45[3].WRKY30 is also associated with activated expression of JA synthesis-related genes LOX, AOS2 and pathogenesis-related (PR)3 and PR10, and increased endogenous JA accumulation under the challenge of fungal pathogens[4].

OsWRKY30 is also involved drought tolerance. OsWRKY30 interacts with the rice mitogen-activated protein kinases(MAPKs), OsMPK3, OsMPK4, OsMPK7, OsMPK14, OsMPK20-4 and OsMPK20-5, and can be phosphorylated by OsMPK3, OsMPK7, and OsMPK14[5].

Mutation

Overexpression of WRKY30 in rice shows enhanced disease resistance to the bacterial pathogen Xoo(fig.1), sheath blight fungus Rhizoctonia solani(fig.2) as well as M. oryzae[3][4].

Overexpression of OsWRKY30 in rice dramatically increased drought tolerance(fig.3). Mutations in the phosphorylation target sites of OsWRKY30 have been shown to cause the loss of function of drought tolerance. That result suggests that OsWRKY30 phosphorylation is important for its biological functions in defense responses to abiotic stress[5].

OsWRKY30 is mostly expressed in rice leaves. It is first induced at 1 h p.i. in cv.Kasalath. Its expression level increased up to maximum by 24 h p.i., decreased slightly by 48 h p.i., produced a second peak around 72 h p.i., and finally declined by 96 h p.i.(fig.4)[6].

As shown in Fig. 5, the WRKY30 transcripts began to accumulate at 1 h and strongly increased during 6–12 h after MeJA treatment, followed by a sharp decrease to the background level at 24 h. Also, the expression of WRKY30 increased rapidly but modestly within 1 h, continued to increase at 6 h, and maintained at a steady level till 24 h after SA treatment[4].
