Annotated Information

Function

OsCPK10 is a member of OsCPKs family including 29 CPKs in rice[1]. Calcium-dependent protein kinases (CDPKs) are improtant calcium sensors icluded in plant responses to pathogen infection. The expression of OsCPK10 was strongly induced following treatment with a Magnaporthe grisea elicitor. Kinase activity assay showed that the functional OsCPK10 protein not only autophosphorylated, but also phosphorylated Casein in a calcium-dependent manner. Overexpression of constitutively active OsCPK10 in Arabidopsis enhanced the resistance to infection with Pseudomonas syringae pv. tomato, associated with elevated expression of both SA- and JA-related defense genes. Similarly, transgenic rice plants containing constitutively active OsCPK10 exhibited enhanced resistance to blast fungus M. grisea. Collectively, rice OsCPK10 is a crucial regulator in plant immune responses, and its overexpression may regulate disease resistance by activating both SA- and JA-dependent defense responses[2].

Protein Structure

OsCPK10 has a variable domain (residues 1-132), a kinase domain (residues 133-396), a junction domain (residues 397-427) and a Calmodulin-like domain (residues 428-599). Four EF-hands were found in the Calmodulin-like domain (residues 438-466, 474-502, 510-538, and 544-572)[2].

Expression

The expression of OsCPK10, in M.grisea elicitor-treated rice calli, shows a rapid activation and peaks at 1 hour, which is earlier than that of OsPAL1 expression. This result indicates that OsCPK10 is an early pathogen-responsive gene[2].

Isolation and sequence analysis

OsCPK10 is isolated by PT-PCR from the total RNA extraction using specific primer pairs and the coding region of OsCPK10 is 1800 bp in length which encodes 599 amino acids. (Primers:forward 5'-ATGGGGAACACGTGCGTC-3' and reverse 5'-TCAAGACATCCTCAAGGCCTC-3'). The PCR products were ligated to a pEASY-T1 vector (TransGen Biotech, China)[2].
