OsLEA4 is a late embryogenesis abundant (LEA) protein gene from rice[1].

Annotated Information

Function

  • OsLEA4 is one of the important components in cellular glasses which may retain water molecules, prevent crystallization of the cellular components, and stabilize desiccation-sensitive proteins as well as plasma membranes during dehydration. OsLEA4 protein enhanced the tolerance of E. coli recombinant to high salinity, heat, freezing, and UV radiation, which suggested that OsLEA4 protein may play a protective role under stressed conditions[1].
  • It is speculated that OsLEA4 in association with other factors has a potential role in protecting cellular components against ROS-induced damage; OsLEA4 might have an additional role as repair mechanisms in eliminating the thymine dimer resulting from the photochemical injury caused by UV radiation[1].

Expression

  • OsLEA4 protein contains a Pfam:LEA_1 domain architecture at positions 1–73 with three α-helical domains and without β-sheet domain. OsLEA4 fusion protein could be expressed effectively and is soluble, which was concordant with the predicted result that OsLEA4 is hydrophilicity.
  • Overexpression of OsLEA4 can improve the tolerance of recombinant E. coli under extreme abiotic stresses including high salinity, heat, freezing, and UV radiation, suggesting that OsLEA4 gene may play role in plant’s ability to adapt to adverse environments. It might be more advantageous to cope with those varying circumstances[1].

Evolution

  • The sequence analysis indicated that the complete ORF of OsLEA4 cDNA is 312 bp and encodes a putative polypeptide of 103 amino acids with a calculated molecular mass of 11.19 kDa. The OsLEA4 polypeptide shows preponderance of Ala (22%), Lys (15%), Glu (9%), His (8%), Thr (8%), and Arg (7%) but depleted in Trp, Cys, Asn, and Phe residues. The OsLEA4 is a basic protein with a theoretical pI of 10.04, which is concordant with the high proportion of positive residues(29%)[1].
  • OsLEA4 belongs to the group 4B. Like most members of the group 4B, there is not any β-sheet domain in OsLEA4. OsLEA4 contains three α-helical domains in the N-terminal “LEA_1” motif[1].

Knowledge Extension

  • Four (11.8%) of the OsLEA genes were found to have alternative splicing, which results in either two (for OsLEA8, OsLEA14, and OsLEA19) or three (for OsLEA7) forms of mature mRNA. This frequency of alternative splicing is much lower than the average (27%) occurred in the rice genome[2][3]. In general, the OsLEA genes contain very few introns. Most (22/34 or 64.7%) of them contain only one intron; seven (20.6%) have two introns; and five (14.7%) have no intron[2]. LEA proteins are late embryogenesis abundant in the seeds of many higher plants and are probably universal in occurrence in plant seeds[4].
  • LEA mRNAs and proteins can be induced to appear at other stages in the plant's life by desiccation stress and/or treatment with the plant hormone abscisic acid (ABA). A role in protecting plant structures during water loss is likely for these proteins, with ABA functioning in the stress transduction process. Presented here are conserved tracts of amino acid sequence among LEA proteins from several species that may represent domains functionally important in desiccation protection[4].
